abstract | The present invention provides methods and adapters for characterizing polynucleotides of interest. The present invention provides a method of characterizing a target polynucleotide, comprising: (a) moving the target polynucleotide through a nanopore, wherein the sequencing terminus of the target polynucleotide comprises a modified portion that enables Nanopore plugging; (b) taking one or more electrical and/or optical measurements as the target polynucleotide moves relative to the pore. The present invention also provides an adaptor for characterizing a polynucleotide of interest, wherein the adaptor comprises a modified moiety that binds to a sequencing terminus of the polynucleotide of interest, the modified moiety clogging the nanopore . The method provided by the present invention avoids a large amount of enrichment of the sequenced library at the trans (Trans) end of the nanopore sequencing device, and further improves the variability of the trans (Trans) end modification. |
