The invention belongs to the field of artificial tissue/organ repair materials, and provides a preparation method and application of a polyvinyl alcohol-agarose hydrogel with a directional microstructure. Using polyvinyl alcohol and agarose as matrix materials, the cross-linking properties of agarose are used to form agarose macromolecular sugar chains, and the flow of the precursor liquid guides the agarose macromolecular chains to generate directionality in the precursor liquid, and in the process of freezing cycle The polyvinyl alcohol crystallizes around the sugar chains of agarose to form a hydrogel with a directional fibrous microstructure. The preparation method does not need to rely on the action of an external physical field, and the preparation of the hydrogel has the advantages of anisotropy; and the preparation process is simple, the repeatability is high, and the equipment requirements are low. The prepared hydrogel has a uniform directional fibrous microstructure, which improves the tensile properties of the material, and can be used to prepare a directional hydrogel of any shape through a syringe.
