The invention relates to a SERS substrate and a preparation method and application. The SERS substrate contains a bottom layer and a surface layer, the bottom layer is a glass flake substrate, and the surface layer is a composite silver nanowire-gold nanorod layer; the silver nanowire-gold nanorod layer is a silver nanowire mesh structure As a base, gold nanorods are attached to a network of silver nanowires. The substrate can realize the quantitative and qualitative detection of three common environmental pollutants, Rhodamine 6g, Crystal Violet and Famex, with detection limits of 10 ‑12 M, 10 ‑10 M and 10 ‑10 M, respectively.
