abstract |
The invention relates to the technical field of plant virus detection, in particular to a method for detecting tomato yellow leaf curl virus based on Northern hybridization. The following steps are included: Step 1. Design primers C4-F and C4-R according to the genome sequence of tomato yellow leaf curl virus, and prepare a tomato yellow leaf curl virus-specific DNA probe with digoxigenin label by PCR method; step 2. Tobacco total RNA extraction and separation of total RNA on agarose gel 3 days after infection with tomato yellow leaf curl virus; step 3. The total RNA isolated in step 2 was transferred to a positively charged nylon membrane by siphoning, and after the transfer was completed The total RNA on the membrane was immobilized under a UV cross-linker; step 4. pre-hybridization, and use the probe prepared in step 1 for hybridization; step 5. washing the hybridization membrane and detecting the hybridization signal. The method overcomes the shortcomings of the existing detection technology, and provides a method for sensitive and specific detection of tomato yellow leaf curl virus. |
