titleOfInvention A method for rapid identification of psychrophilic extracellular thermostable protease species
abstract The present invention relates to a method for rapidly identifying psychrophilic bacteria extracellular heat-resistant protease species, comprising the following steps: (1) centrifuging psychrophilic bacteria fermentation broth to prepare fermentation broth samples; (2) configuring SDS-PAGE containing protein substrates gel; (3) mix the fermentation broth sample with the protein loading buffer and load the sample; (4) electrophoresis; (5) take out the gel and perform gel elution with pre-cooled eluent; (6) The gel was incubated in incubation buffer; (7) Coomassie brilliant blue staining. Compared with the prior art, the method of the present invention can rapidly characterize the extracellular heat-resistant protease species of psychrophilic bacteria, can distinguish the heat-resistant protease species of different molecular weights well, and can more accurately characterize the heat-resistant protease species than the traditional method. The tedious protease separation and purification steps are omitted, the repeatability is good, the sensitivity is high, and it can be used for the detection of extracellular heat-resistant proteases of different psychrophilic bacteria, and has a wide application space.
