A method for preparing an ACE2 siRNA drug for delivering shRNA by targeting ACE2, screening target siRNA from coronavirus conserved genes or microsatellites, synthesizing a small hairpin shRNA with a loop from two complementary positive and antisense siRNA strands, and synthesizing a small hairpin shRNA with a loop. The receptor binding domain RBD is ACE2 or penetrating peptide ACE2, and then ACE2 is connected to the positive and antisense strands of shRNA respectively to synthesize ACE2siRNA composed of shRNA region and ACE2 region, in which bivalent ACE2 neutralizes RBD and ACE2 siRNA. The role of targeted delivery of shRNA, the ACE2-bridged "shRNA-ACE2-RBD-virus" complex allows shRNA to enter target cells with virus infection, which not only avoids the side effects of non-specific delivery of shRNA to uninfected cells, but also reduces the risk of infection. It can produce anti-mutant strains and neutralize viruses with ACE2. In addition, the immune adjuvant effect of ACE2 siRNA and shRNA synthesized by two molecules of ACE2 and one molecule of shRNA can make ACE2 have stronger antigenicity and stimulate the body to produce anti- ACE2 in turn has antiviral effects. |
