abstract | The invention provides a method for preparing a DchCHS1 gene overexpression vector of Chinese dianthus. The method comprises the following steps: extracting total RNA from leaves of Chinese dianthus Dianthuschinensis, and reverse transcribing it into cDNA; using cDNA as a template, performing PCR amplification of DchCHS1 gene to obtain After the PCR product was recovered, the pMD19‑T‑DchCHS1 plasmid was prepared, the pMD19‑T‑DchCHS1 plasmid was digested, and the DchCHS1 fragment was ligated to the digested pBI121‑GUS to prepare the pBI121‑DchCHS1‑GUS plasmid. The GUS plasmid was transformed into DH5α competent cells. After PCR reaction verification and sequence comparison, the cloned pBI121-DchCHS1-GUS recombinant plasmid was successfully cloned, which was named as the Chinese dianthus DchCHS1 gene overexpression vector. An application is also provided, wherein the DchCHS1 gene overexpression vector of Chinese dianthus is prepared into an infection solution, which is used to infect the flower buds of Chinese dianthus, enrich the color of Chinese dianthus petals, and improve the CHS enzyme activity and flavonoid content of the Chinese dianthus petals.
