abstract | The invention discloses a preparation method of oxybutynin hydrochloride (chemical name: α-cyclohexyl-α-hydroxy-phenylacetic acid-4-diethylamino-2-butynyl ester hydrochloride), comprising the following steps: α-Cyclohexyl-α-Hydroxyphenylacetic acid and 3-chloropropyne are esterified with 3-chloropropyne under a suitable solvent and basic catalyst to generate an intermediate: α-Cyclohexyl-α-Hydroxyphenylacetic acid Propargyl ester, then Mannich reaction with 36% formaldehyde solution and diethylamine to generate oxybutynin, and finally salify with hydrochloric acid to generate oxybutynin hydrochloride. The reaction routes of the invention are relatively simple, do not involve reactions under harsh chemical reaction conditions, do not involve high-risk chemical reaction processes, and are suitable for industrial production. |
