The present invention relates to the field of molecular biology, in particular to an IL27-presenting stably transfected cell and a construction method thereof, an IL27-presenting engineered exosome and a preparation method and application thereof, wherein the steps of the construction method for IL27-presenting stably transfected cell are S100 , inserting the IL27 gene sequence into the gene sequence of the exosome membrane protein directionally to obtain a fusion protein expression vector; S200, co-transfecting the fusion protein expression vector and packaging plasmid prepared by the S100 into 293T cells to obtain a lentivirus; S300, The stably transfected cells are obtained by infecting the mesenchymal stem cells with the lentivirus prepared by the S200. The present invention uses mesenchymal stem cells to construct engineered exosomes to present and deliver interleukin 27 (IL27), so that IL27 is directionally expressed on the exosome membrane of mesenchymal stem cells, thereby increasing the local concentration of IL27 action and prolonging IL27 half-life of action. |
