In the present invention, a fluorescent protein gene is in situ connected to the 3' end of the fungal target gene by means of homologous recombination gene manipulation mediated by Agrobacterium, so that the target gene and the fluorescent protein gene are co-expressed in the form of fusion protein (target protein-fluorescent protein). The subcellular localization of the target protein is analyzed by fluorescence observation and its expression is quantified; the fungal fluorescent fusion protein in situ gene expression technology disclosed in the present invention reduces the uncertainty of random insertion and improves the accuracy of the experimental results.
