abstract | The present invention relates to a method for identifying and screening human embryonic bone marrow-derived mesenchymal stem cells, a combination of related detection agents, a kit and related applications. The present invention comprehensively screened human embryonic BM nucleated cells (BMNCs) at single-cell resolution, using two complementary strategies (STRT and 10X genomics) for scRNA‑seq analysis, identified LIFR + PDGFRB + as MSCs specific markers. The present invention also finds that LIFR + PDGFRB + CD45 - CD31 - CD235a - MSC can effectively rebuild the hematopoietic microenvironment in vivo. The present invention also identifies a subset of unipotent osteoprogenitor cells expressing TM4SF1 + CD44 + CD73 + CD45 - CD31 - CD235a-, which have osteogenic potential. Therefore, the present invention identifies two types of human embryonic bone marrow MSCs, and the present invention will deepen the understanding of human embryonic bone marrow-derived MSCs and further promote the understanding of the clinical application of MSCs. |
