abstract |
Compositions and methods for modifying genomic DNA sequences are provided. The method creates double-strand breaks (DSBs) at predetermined target sites in the targeted DNA sequence, resulting in mutations, insertions and/or deletions of the DNA sequence at one or more of the targeted sites. Compositions include a DNA construct comprising a nucleotide sequence encoding a Cpf1 protein operably linked to a promoter operable in a cell of interest. DNA constructs can be used to direct genomic DNA modifications at predetermined locations. Described herein are methods of using these DNA constructs to modify genomic DNA sequences. Furthermore, compositions and methods for modulating gene expression are provided. The composition includes a DNA construct comprising a promoter operable in a cell of interest operably linked to a nucleotide sequence encoding a mutant Cpf1 protein that abolishes the ability to generate DSB, optionally linked to regulate transcription active domain. This method can be used to up-regulate or down-regulate gene expression at predetermined genomic loci. |
