titleOfInvention | Extraction of Fumarase from Saccharomyces cerevisiae |
abstract | The invention discloses a new process for separation and purification of fumaric acid enzyme; the invention is realized as follows: the suspension of Saccharomyces cerevisiae cells is suspended in 100 mol/L potassium phosphate buffer with pH of 7.5 according to the mass fraction of 4 to 10, respectively, Cells were disrupted with a bead mill mixer and ultrasonic ice bath for 30 min, the suspension was centrifuged at 4000 rpm for 5 min, the supernatant was taken to adjust the pH to 7.5, the precipitate was discarded, and the supernatant was added to acetone and allowed to stand at low temperature. ; Its activity is measured by the reduction of malic acid per unit time, and the increase of absorbance at 330nm wavelength is measured by UV-visible spectrophotometer. It is characterized in that the ultrasonic ice bath method is adopted when the cells are broken, and the enzyme protein existing on the subcellular structure can be separated by the cavitation of the ultrasonic wave, thereby improving the separation and purification efficiency. Compared with the ordinary bead milling and stirring purification method, it is found that the fumarate obtained by the ultrasonic ice bath breaking cell method has high yield and high activity. |
