abstract | The invention discloses a novel method for rapid qualitative screening and accurate quantitative analysis of ketamine, which can rapidly quantitatively analyze KET in human blood, urine and environmental samples by using the competition mode of the ketamine hapten-coated microplate. The competitive quantification equation obtained by this method has a linear correlation coefficient of 0.992, a linear range of 0.01μg/mL~0.5μg/mL, and a detection limit of 0.1 pg/ml. The specific results show that the cross-reaction rates of the interfering substances are lower than 10%. The recoveries of low, medium and high concentration spiked samples were 96%~117%. This method and classical GC-MS/MS simultaneously detect real samples, and the relative deviation of quantitative results is less than 10%. Moreover, compared with the GC-MS/MS method, it is found that the LOD value of this method is four orders of magnitude lower than that of the classical GC-MS/MS method in terms of sample pretreatment and economic investment. Fast, portable and sensitive qualitative and quantitative analysis of ketamine can be achieved. |
