The invention discloses a D-amino acid oxidase mutant, engineering bacteria and applications, and belongs to the field of biotechnology. The D-amino acid oxidase gene is derived from Rhodotorula mucilaginosa, and its nucleotide sequence is shown in SEQ ID NO: 1, and its amino acid sequence is shown in SEQ ID NO: 2. The mutant is obtained by single mutation or multi-site mutation at the 54th, 58th, 213th, 223rd and 225th positions of the amino acid sequence shown in SEQ ID NO: 2.
