The invention provides a method for simultaneously enhancing and inhibiting multiple key genes in the synthesis of 7-dehydrocholesterol in Saccharomyces cerevisiae, belonging to the technical field of bioengineering. The present invention achieves the purpose of simultaneously expressing and inhibiting 13 P450 enzymes through the dCpf1-VP activation system and the dCas9-RD inhibition system, and constructs an engineering yeast with high production of 7-dehydrocholesterol, so that the output of 7-DHC reaches 464mg/L. |
