abstract The invention discloses a simple and rapid method for determining the lignin content in lignocellulosic biomass. The enzymatic hydrolyzed lignin is firstly dried, ground into fine powder, then dissolved in [Emim][OAc] ionic liquid, and added with two Methyl sulfoxide was diluted to prepare a standard solution, and the absorbance was measured at 285nm with an ultraviolet spectrophotometer, and a standard curve between the absorbance and the lignin concentration was made to obtain the ultraviolet absorption coefficient ε; the lignocellulosic biomass was ground and dried. Dissolve in [Emim][OAc] ionic liquid system, add diluent for dilution, control the ultraviolet absorbance value of the diluted sample solution under ultraviolet light with a wavelength of 285nm to be 0.2‑1; substitute the ultraviolet absorbance value into calculated in the formula. The invention has the characteristics of simple operation, few procedures, easy availability of reagents, environmental protection in the process, high sensitivity, few samples required, wide application range and more accurate determination results.
