abstract The invention belongs to the field of biotechnology, and in particular relates to a method and application for filtering methylation sequencing data. The methylation sequencing data filtering method provided by the invention can increase the conversion efficiency of unmethylated C bases into U bases in the bisulfite-treated DNA from 98.81% to 99.44%. On this basis, the application of the data filtering method of the present application can accurately assess the methylation level of the sequence and obtain methylation haplotypes with high quality, high accuracy and reliability. The experimental data show that using the methylation haplotype obtained by the methylation sequencing data filtering method provided by the present invention can well distinguish the urine samples of bladder urothelial carcinoma patients and normal people (AUC=0.94) and Urine samples from patients with muscle-invasive bladder urothelial carcinoma versus non-muscle-invasive bladder urothelial carcinoma (AUC=0.967).
