abstract | The invention provides a colloidal gold labeling method with improved sensitivity, application and product thereof, and relates to the technical field of immunochromatography. In the colloidal gold labeling method provided by the present invention, the protein to be labeled treated with a protein activator is mixed with a colloidal gold solution to carry out labeling reaction to obtain a gold-labeled protein. Among them, protein activators include non-ionic surfactants, carbohydrate protective agents and reducing agents. Through the synergistic cooperation between the components, the interaction between the gold-labeled conjugates is effectively reduced, aggregation is avoided, and the gold-labeled binding agent is improved. The stability of the conjugate protects the activity of the protein; the pH of the colloidal gold solution is lower than the isoelectric point of the protein to be labeled by 0.3-0.8, and the protein is labeled under this condition, and the formed gold-labeled conjugate has good stability and high labeling efficiency. The method is easy to operate, the prepared gold-labeled conjugate has high labeling efficiency and good stability, and the prepared kit has high sensitivity and is not easy to miss detection. |
