abstract The invention discloses an in vitro hepatotoxicity surrogate model constructed by HepaRG and HepG2 and a construction method thereof. The model is obtained by culturing and differentiating two kinds of human hepatocytes under conventional conditions. After differentiation, HepaRG cells have enhanced metabolic capacity and increased sensitivity to compounds. The established model can be used to detect a variety of hepatotoxic compounds, and can also be used to explore the toxicity mechanism of known hepatotoxic drugs. After in vitro verification, it is shown that the hepatotoxicity markers screened by the present invention are suitable for the evaluation of the hepatotoxicity of food contaminants, and their expression levels can be used for the classification of hepatotoxicity grades. The in vitro hepatotoxicity surrogate model constructed by HepaRG and HepG2 of the present invention can be applied in the safety evaluation of hepatotoxic drugs. Compared with the traditional liver function index, the measurement result of the invention is more stable, and has been verified by animal experiments, which makes up the blank of the previous in vitro research on the hepatotoxicity of food contaminants.
