The invention relates to a method for measuring milk fat. On the basis of analytical technical means, a method for separating triglyceride and phospholipid in a sample to be tested based on multi-heart-cutting-two-dimensional liquid phase is proposed for the first time. Chromatography (HILIC) combined with reversed-phase chromatography for the separation of complex components, combined with post-column light derivatization and ion mobility analysis, and time-of-flight mass spectrometry quantification, can identify all triglycerides and triglycerides in the test sample through comprehensive analysis. Phospholipids, including information on fatty acid composition, double bond position, cis-trans isomerism at Sn-1, 2, 3 positions of triglyceride and phospholipid R1, R2 positions. |
