abstract |
The present invention provides a method for improving the correct pairing rate of heavy and light chains during the preparation of bispecific antibodies. The method includes eliminating natural interchain disulfides by amino acid substitution on the CH1-CL domain of one Fab arm. bond, while forming an engineered interchain disulfide bond by amino acid substitution. The method of the present invention further comprises the step of reversing the charge of a pair of amino acids of the CH1-CL domain of the Fab arm by amino acid substitution. The method of the present invention can significantly improve the rate of correct pairing of heavy and light chains in bispecific antibody production, and is applicable to various antibody types. |
