abstract The invention discloses an oligonucleotide separation and purification method, which comprises the step of subjecting an oligonucleotide sample to be separated to non-denaturing polyacrylamide gel electrophoresis so as to separate the oligonucleotides, wherein the concentration of polyacrylamide in the gel is 20-28%. The invention also discloses an oligonucleotide purification method, which comprises the following steps: (1) isolating an oligonucleotide sample using the oligonucleotide isolation method described above; (2) recovering the separated oligonucleotides. The method for separating and purifying the oligonucleotide provided by the invention has higher resolution and better separation effect, and can save the process time.
