Heater for thermal cycling of PCR amplification. The heater includes: a thermal diffusion layer having a reaction surface for transferring heat to the reaction unit; a heater track support layer having a rear surface for cooling; supported on the heater track support layer and the heat a conductive main heater track between the diffusion layers; and a four-terminal electrical contact to the main heater track adapted to provide a resistance for driving the main heater track while sensing the main heater track electrical connection. The lateral dimension of the reaction surface is greater than the thickness H of the heater such that the reaction surface area A>H 2 . |
