abstract | The invention relates to a method for efficiently detecting anti-retinal antibodies (ARAs) in serum, which comprises the following steps: (1) extracting venous blood of a clinical suspected AIR patient, and centrifuging to obtain serum of the patient; (2) detecting the number of antibody positive bands, namely the number of ARAs, in serum of a patient by a western blot method by taking retinal protein extracted from normal human retinal tissue homogenate as a reaction substrate; (3) the known human recombinant protein antigen is used as a reaction substrate, and the types of ARAs existing in the serum of a patient are detected and confirmed by a western blot method; (4) detecting ARAs in the serum of a patient by adopting an immunofluorescence method, and positioning the ARAs in a normal human retina tissue slice to obtain the positioning of the ARAs in the serum of the patient in a retina tissue; (5) and (3) determining the quantity of serum ARAs, the types of serum ARAs and the positioning of the ARAs in the retinal tissue according to the detection results of the serum ARAs of the patients obtained in the steps (2), (3) and (4), and establishing the serological index of the clinical suspected AIR patient. The detection method of the invention has high sensitivity and specificity, and is suitable for clinical early detection. |
