abstract | The invention discloses a kit and a method for detecting DNA fragments of active sperms, wherein the kit comprises: a Pi detection reagent which is a complex of a fluorescent substance and silver, and when Pi is present, Pi substitutes for silver in the complex so that the quenched fluorescent substance returns to fluorescence; an acidizing fluid which comprises hydrochloric acid, sodium chloride and triton X-100; a buffer comprising sodium chloride, ethylenediaminetetraacetic acid, tris (hydroxymethyl) aminomethane hydrochloride, and citric acid; and AO staining solution. In the present invention, a carbon quantum dot that should be quenched due to formation of a complex with silver is used as a Pi detection reagent, and when Pi is present, it returns to green fluorescence, thereby enabling specific recognition of a motile sperm; based on a flow cytometer and matched with a sperm chromatin structure detection method based on an acridine orange dye, the sperm motility rate and the DNA fragmentation rate of the moving sperm can be detected, the quality of the sperm can be more directly reflected, and an evaluation index with higher reference value is provided. |
