abstract | A preparation method and application of D-psicose 3-epimerase immobilized enzyme. The invention provides nucleotide sequences of an interacting polypeptide pair Spycatcher/SpyTag and a snopCatcher/snopTag for preparing a D-psicose-3-epimerase immobilized enzyme, a method for preparing the D-psicose-3-epimerase immobilized enzyme by using the four polypeptides, and a method for producing the D-psicose by using the immobilized enzyme. The D-psicose-3-epimerase immobilized enzyme prepared by the method provided by the invention simultaneously keeps the characteristics of high enzyme activity and high stability, and the enzyme activity is not obviously attenuated after repeated use for multiple times. Meanwhile, the preparation of the D-psicose-3-epimerase immobilized enzyme by using the polypeptide pair provided by the invention does not need glutaraldehyde (crosslinking) and other toxic chemical reagents in the process, does not relate to expensive reagents and complex equipment, and has no toxicity and low cost in the whole preparation process. |
