abstract The invention relates to a construction method and application of a glycine riboswitch gene regulation circuit; through mutation ON a wild type glycine riboswitch element sequence, glycine-ON mutants with improved performance, increased glycine response range and increased amplitude value and glycine riboswitch mutants with changed conformation are screened out from a library at high flux, so that the expression regulation of genes can be realized. The invention simultaneously converts wild glycine-ON into glycine-OFF riboswitch and increases the amplitude value of the riboswitch. The invention also provides a method for screening glycine mutants with improved performance and changed conformation, and an expression vector and a host cell containing the mutants. The invention discloses a glycine riboswitch gene regulation circuit construction, performance improvement and conformation change mutant, which is an effective mutant based on a glycine riboswitch circuit and performance improvement reported for the first time in escherichia coli at present.
