abstract The invention discloses a device for measuring the net carbon fixation rate of unicellular photosynthetic microorganisms, which comprises a gas storage tank; the glass tube is hermetically connected with the air outlet of the air storage tank through an air inlet and is provided with a plurality of air outlets; the three-way valve is connected with the air outlet, a needle head is arranged at the outlet of the three-way valve and used for being connected with the brine bottle, and rubber plugs are arranged at the outlets of the needle head and the three-way valve; and the gas chromatograph is used for measuring the solution in the saline bottle. Also included is a method of measuring net carbon sequestration rate of a unicellular photosynthetic microorganism, S1: inoculating; s2: culturing; s3: connecting a ventilation device; s4: ventilating; s5: carrying out closed culture; s6: sampling; s7: gas chromatography analysis; s8: counting the cells; s9: and (4) calculating. The measuring method has the advantages of simple operation steps, measurement time saving, rich and easily-obtained raw materials such as the used three-way valve, the used syringe, the used saline bottle, the used rubber tube and the like, convenience in assembly, extremely low cost and suitability for measurement application in a laboratory.
