abstract | The invention discloses a method for detecting methotrexate and folic acid metabolic pathways, which is characterized by comprising the following steps: (1) dissolving a standard substance in an ammonium hydroxide solution, mixing, and diluting by using a bovine serum albumin solution to obtain a standard solution; (2) respectively placing 50ul of standard solutions with different concentrations and serum samples in a centrifuge tube, adding 200ul of methanol solution containing an internal standard, carrying out vortex oscillation, carrying out high-speed centrifugation at 13000rpm, and sucking 100ul of supernatant for detection and analysis; (3) performing liquid chromatography tandem mass spectrometry (4) on the supernatant obtained in the step (2) to obtain a linear regression equation, namely a standard curve; (5) and obtaining the concentration of the object to be detected in the serum sample according to the standard curve. The invention has accurate and sensitive measurement and high flux, can be used for simultaneously researching the pharmacokinetics of the methotrexate and the folinic acid in the high-dose methotrexate treatment process, and can be used for optimizing the rescue treatment of the high-dose methotrexate and the folinic acid in individual patients. |
