abstract |
The invention discloses a construction method and application of a Iridaceae plant eleutherine rhizome VIGS silencing system. The invention uses tobacco rattle virus TRV as the vector of the VIGS system, uses the phytoene dehydrogenase gene of the red onion as the target gene, and successfully creates the VIGS silencing system suitable for the red onion by using a vacuum infiltration treatment method; the system is applied to the research on the functions of genes EbMYB5 and EbMYB52 related to the red onion pigment synthesis, the expression levels of the EbMYB5 and EbMYB52 genes in the infected silent treatment group sample blades are obviously reduced, and the expression levels of key genes related to the pigment synthesis path show obvious changes of different degrees, which shows that the red onion VIGS silencing system can be successfully applied to the red onion gene function verification. The ViGS silencing system constructed by the invention has important significance for researching the gene function of the red onion. |
