The invention provides an amplification primer of a thymidine kinase gene of paulownia arbuscular phytoplasma, and two genotypes are obtained by amplification; constructing a prokaryotic expression vector of thymidine kinase of paulownia arbuscular phytoplasma, and performing induced expression and purification to obtain 6 XHis/PaWB TDK-1 fusion protein and 6 XHis/PaWB TDK-2 fusion protein; rabbit polyclonal antibody is obtained by immunizing rabbit with fusion protein as immunogen; the polyclonal antibody can generate immunoreaction with both PaWB TDK-1 protein and PaWB TDK-2 protein; by means of immunoblotting, immunofluorescence microscopy and immunoelectron microscopy, the polyclonal antibody is proved to be better used for detecting the paulownia arbuscular phytoplasma and thymidine kinase expression thereof. The invention lays a foundation for further researching the multiplication pathogenesis of phytoplasma and provides a new technical means for detecting the phytoplasma of paulownia arbuscular branches in agriculture and forestry production. |
