The invention discloses a Taiwan loach elovl5 gene, the nucleotide sequence of which is shown in SEQ ID No. 1. The invention also discloses application of the elovl5 gene in molecular breeding for improving the content of highly unsaturated fatty acid of Taiwan loaches and a molecular breeding method for improving the content of highly unsaturated fatty acid of Taiwan loaches, and the method utilizes a CRISPR/Cas9 system to knock out the elovl5 gene to obtain an elovl5 gene knock-out mutant of the Taiwan loaches. The experimental result shows that compared with the wild type, the knockout type has obvious difference in the content of both EPA and DHA in vivo, compared with the wild type, the knockout type has obvious increased content of EPA and DHA, and the total content of DPA, EPA and DHA is obviously increased. |
