The invention relates to the application of a replicating human type 3 adenovirus as an expression vector in preparing a pig vaccine. The present invention is the first attempt to use replicative human adenovirus type 3 as an expression vector to express foreign proteins such as viral antigens to prepare vaccines for pigs. We constructed various types of replicative human adenovirus vectors, including human adenovirus types 3, 4, 7, and 55, inserted the foreign gene expression box EGFP in the E3 region, and used the four replicative adenovirus vectors. The virus vector infects pig cells, and it is found that the human adenovirus type 3 vector can replicate the vector genome more efficiently in pig cells, and the expression efficiency of foreign genes is higher, and the human type 3 adenovirus vector produces infectious progeny in pig cells. The titer was significantly lower. |
