abstract |
The invention provides a PPIs detection platform and a detection method based on a CRISPR-Cas12a sensing system, which are used for monitoring the interaction of p53-MDM2 and evaluating the curative effect of an inhibitor. The detection method comprises the following steps: performing probe design on the Cas-PPIor and verifying the feasibility of the Cas-PPIor; Cas-PPIor was used for p53-MDM2 interaction studies and inhibitor efficacy evaluation; Cas-PPIor is used for intracellular protein interaction detection. The invention develops a PPIs detection platform based on a CRISPR-Cas12a sensing system, and can realize effective monitoring of the interaction process of p53-MDM2 and evaluation of the inhibition effect of a small molecule antagonist. |
