titleOfInvention Material taking method suitable for DRG (dorsal root ganglion) of rat
abstract The invention belongs to the technical field of tissue sample acquisition, and discloses a material taking method suitable for rat dorsal root ganglion DRG, which comprises the following steps: taking dead rats to determine the position of an iliac crest, and cutting off and separating spines; putting the separated spine into hydrogen peroxide for washing and soaking for one minute; finding the L5 vertebral body from the cross section of the removed lumbar spine segment; respectively shearing off the muscles on the horizontal planes at the two sides of the vertebral body; separating the converged spinal nerves and muscle tissues in sequence, taking out the dorsal root ganglia and the central ends and the peripheral ends of the two ends of the dorsal root ganglia, and only keeping the dorsal root ganglia; putting the dorsal root ganglion into a centrifuge tube, temporarily putting into liquid nitrogen, and transferring into a refrigerator at the temperature of minus 80 ℃ for storage. The invention can quickly position the spinal ganglia and ensure that the dorsal root ganglia are not polluted by bloodstains, thereby providing high-quality tissue samples for histological and morphological experiments. The invention has the advantages of less time consumption, simple and convenient operation, more complete back root ganglion and no pollution by bloodstain.
