abstract The invention discloses a plastic embedding method for simultaneously keeping multicolor fluorescence signals and an ultrastructure and application thereof, wherein the specific process comprises the following steps: pretreating a fluorescence-labeled biological tissue sample; putting the pretreated biological sample into gradient ethanol and embedding medium for infiltration; polymerizing the sample subjected to gradient treatment at-20-4 ℃ by using an embedding medium added with a low-temperature polymerization promoter to obtain an embedded sample, and achieving the purpose of stably polymerizing the biological tissue sample at low temperature by adding the low-temperature polymerization promoter. The plastic embedding method capable of simultaneously keeping the multicolor fluorescence signal and the ultrastructure can stably realize plastic embedding of a whole brain sample at the temperature of-20-4 ℃, cutting at a submicron level, well keeping the ultrastructure, well keeping the multicolor fluorescence signal and not increasing the background light of biological tissues, can be applied to fluorescence microscopic imaging, and has certain potential application to electron microscope imaging.
