abstract The invention provides a preparation method and application of an adeno-associated virus capsid protein conserved region polyclonal antibody, through DNAMAN software analysis, sequences homologous with continuous 7 or more than 7 amino acids in AAV1, AAV2, AAV3, AAV4, AAV5, AAV6, AAV7, AAV8, AAV9 and AAV10 capsid protein amino acid sequences are spliced in sequence, the obtained sequence is AAV capsid protein conserved region sequence, DNA sequence of coding capsid protein conserved region can be determined corresponding to conserved region amino acid sequence, codon optimization is carried out on the obtained DNA sequence according to the characteristics of host bacteria, the optimized DNA sequence is synthesized and constructed to pET-30a prokaryotic expression vector, pET-30 a-AAV-conserved region protein plasmid is obtained, the DNA sequence is SEQ ID NO.1, and the amino acid sequence is SEQ ID NO. 2. The polyclonal antibody of the AAV capsid conservative region protein is successfully prepared, all serotype AAV is identified, and the AAV capsid protein which is transfected and expressed in human eukaryotic cells is specifically identified and combined.
