abstract The invention relates to the technical field of microbial fermentation strain preparation, and discloses a passage method of a Streptomyces tenebrarius strain, which comprises Streptomyces tenebrarius spores, wherein the Streptomyces tenebrarius spores stored in a glycerin pipe at the temperature of-20 ℃ are taken. According to the passage method of the Streptomyces tenebrarius strain, the culture medium of the culture dish has uniform thickness and uniform distribution of nutrient components, so that single colony spores on the culture medium are in the same nutrient condition, sufficient nutrient components are provided for spore growth, and the phenomenon that the normal growth of the spores is influenced due to insufficient absorption of part of the spores on the slant culture medium due to inconsistent thickness is avoided. Through diluting the coating to glycerine tube spore, single spore can separate in the culture dish culture medium and grow into the single colony, observes the form of every colony easily to will show the decline and the inconsistent colony separation of most colonies, keep the good single colony spore of phenotype, avoided on the inclined plane culture medium can not separate the shortcoming of bad spore.
