abstract The present invention provides a set of universal circularization chaperone sequences for different high-throughput sequencing platform libraries, a kit thereof and a circularization method, including: adding three sets of single-end, double-end, and double-ended sequences to the ligation buffer (splint buffer) The end-tag library and the splint oligo fragment of the complementary part of the Illumina library adapter can simultaneously circularize single-stranded DNA for different types of libraries; and add promoters to the universal circularization ligation buffer system to improve the circularization efficiency . The invention is convenient, flexible and highly compatible, and solves the problem of using corresponding cyclization reagents for different types of libraries.
