The present invention designs the regulated expression of a chimeric antigen receptor (CAR) in a lentiviral vector. The CAR contains a HOOK-binding domain that interacts with HOOK, preferably encoded by the same lentiviral vector, which prevents the CAR from being properly processed and released to the cell membrane. The present invention includes carriers, methods of making carriers, and methods of using carriers, including medical uses. The vector can be used for administration to humans to induce an immune response and treat cancer and tumors. |
