The present invention utilizes the characteristics of many repeating sequences in the IGHG region of the porcine immunoglobulin heavy chain, and realizes multiple cleavage at multiple sites in the IGHG region by using the CRISPR-Cas9 technology of combining the four gRNAs shown in Sequence 2 in the sequence table with nucleic acid molecules Finally, the simultaneous knockout of the IGHG region up to 110kb in the constant region of the immunoglobulin heavy chain in pigs that affects the immunogenicity of the antibody is achieved, which lays the foundation for the establishment of human immunoglobulin transgenic pigs.
