The invention discloses a method for separating and enriching microorganisms on the surface of alcoholized tobacco leaves. The method comprises the following steps: (1) mixing alcoholized tobacco leaves with phosphate buffer, quartz sand and glass beads, and shaking the mixture at low temperature to obtain a microbial eluate on the surface of tobacco leaves; (2) filtering the microbial wash obtained in step (1) Deliquoring, placing the filtered eluate in the first container, fixing the microporous membrane to the mouth of the first container, and inverting the first container into the second container to obtain the separated microorganisms ; Collect the microporous membrane in step (2) in a third container, add steel beads and phosphate buffer, shake, elute and centrifuge to obtain enriched microorganisms. The method is fast and efficient, and is especially suitable for the separation and enrichment of surface microorganisms in alcoholized tobacco leaves.
