abstract | The invention discloses a screening method and an immobilization method for petroleum degrading bacteria, and an application of the immobilized bacterial agent. The method includes step (1) inoculating oilfield sludge into an enriched medium for culturing to obtain an enriched medium suspension (2) the enrichment medium suspension is inoculated on the LB solid medium and cultivated to obtain a single bacterial colony; (3) the single bacterial colony is inserted into the inorganic salt medium, and the removal test is carried out to obtain a high target bacteria of the oil removal rate. (4) Compound the target strains and insert them into the inorganic salt medium for removal test; the enrichment medium includes peptone, yeast extract powder, and NaCl; the inorganic salt medium includes NH 4 NO 3 , KH 2 PO 4 , K 2 HPO 4 , MgSO 4 ยท7H 2 O, FeCl 3 , CaCl 2 . Through the screening method, high-efficiency petroleum-degrading bacteria containing six kinds of petroleum-degrading bacteria can be obtained by screening and compounding, which has a good removal effect on petroleum hydrocarbons. |
