abstract The invention discloses a method for creating a new broccoli variety with high sulforaphane content and application of the method for cultivating the new broccoli variety, which comprises the following steps: myrosinase gene bolmyr -01 coding sequence synthesis; with restriction enzyme sites bolmyr -01 amplifying; constructing an overexpression vector; genetic transformation; over-expression bolmyr -01 broccoli positive line screening; over-expression bolmyr -01 inbred homozygosis of broccoli; over-expression bolmyr -01 determination of the content of the broccoli pure line sulforaphane and evaluation of the agronomic characters. The results show that the overexpression is carried out in the different germplasm materials of broccoli under the same growth conditions bolmyr The-01 does not affect the shape and the yield of the flower ball, but can obviously improve the content of the sulforaphen in the germplasm material. Over-expression bolmyr The content of sulforaphane in-01 broccoli seed materials can reach 5-10 times of that of a control. The method disclosed by the invention has important values for efficiently, quickly and directionally creating the new broccoli germplasm, enriching the broccoli germplasm resources and cultivating the new broccoli variety.
