abstract |
The invention relates to the field of biomedicine, in particular to a gene expression cassette, a lentiviral vector and its application in the treatment of beta thalassemia. The gene expression cassette includes a sequentially connected promoter, a beta globin gene, an intron-BCL11A-shRNAmir and a polyadenylation signal; the promoter is an erythroid cell-specific type II promoter. The gene expression cassette constructed by the present invention can specifically up-regulate the expression of β-globin and γ-globin in erythroid cells, the two mechanisms act synergistically, and the therapeutic effect is significantly enhanced. |
