The invention discloses a method for separating and purifying phytol from red hair algae, which comprises the following steps: adding ethanol into the powder of Haematococcus, extracting at room temperature for 3 times, filtering, mixing filtrates, and concentrating under reduced pressure to obtain extract; suspending the crude extract by 10 times of volume of water, extracting for three times by using petroleum ether in an equal volume ratio, combining extract liquor, and then concentrating under reduced pressure to obtain a petroleum ether phase; adding petroleum ether into the petroleum ether phase for redissolution, preparing by adopting a rapid preparative chromatography, eluting at a flow rate of 50mL/min, linearly eluting by using the petroleum ether and ethyl acetate, collecting 80% of petroleum ether components, and removing the solvent to obtain a phytol crude product; and carrying out PTLC (polymer PTLC), spotting, developing, scraping and sample recovery on the phytol crude product to obtain a phytol pure product. Therefore, phytol can be efficiently prepared, the purity of the obtained phytol is high, impurities are few, the loss of the effective activity of the phytol is avoided, and the stability of the phytol is facilitated. |
