As an important member of the miRNA family, miRNA-21 has been widely noticed as a tumor biomarker of breast cancer. Because of its short sequence, low expression level in cells and high sequence similarity with other family members, there still exists a certain difficulty and challenge in clinical application for highly sensitive detection of miRNA-21. The invention provides a novel metal organic framework ratio type fluorescent probe based on gold nanoclusters, which is characterized in that a metal organic framework material with large specific surface area and high porosity is used as a carrier, the gold nanoclusters are generated by an in-situ synthesis method, and a designed and synthesized double-stranded probe is assembled on the surface of the carrier. The probe constructed by the invention is used for detecting miRNA-21, overcomes the defects of low detection sensitivity, high background signal and the like of the traditional method, solves the problems of complex living body and cell imaging technology, high price and the like, and provides a new strategy and method for living body and cell detection and in-situ imaging. |
