The invention designs specific primers, uses overlapping PCR amplification technology to connect the fiber protein 1 and fiber protein 2 genes of FAdV-4 through the 2A peptide gene to form a fusion gene fibers2A, and then insert the fusion gene fibers2A into pCDNA3.1 eukaryotes Expression vector, construct recombinant plasmid pC-fibers2A, transform the recombinant plasmid pC-fibers2A into Escherichia coli DH5α, and extract the recombinant plasmid after expanding and culturing, which is a DNA vaccine. The recombinant plasmid was used in immunization experiments, and the results showed that the DNA vaccine could immunize SPF chickens to produce antibodies, reduce morbidity and mortality, and improve the effect of immunization prevention.
