The invention discloses a preparation method of chitosan microspheres for cell immobilization and drug delivery, and relates to the field of preparation of chitosan microspheres. The invention deacetylates the chitin by combining a complex enzyme and a microwave induction process technology, adopts tetrasodium glutamate diacetate or citric acid to carry out heavy metal green chelation on the chitosan, and further utilizes a carbonate dissolution promotion technology to improve the extraction and removal capacity of the chelating agent on residual metal ions; and hydrolyzing and separating trace protein and lipid in the crude chitosan by selecting proper biological enzyme to obtain refined chitosan, performing enzymatic molecular modification on the refined chitosan by using transglycosylation or oxidase, and finally preparing the chitosan microspheres by using an environment-friendly spray drying method. The chitosan microspheres prepared by the method are smooth in shape, uniform in size, 1-250 mu m in particle size and core-shell in structure, and the application of chitosan in the fields of medicine, food and the like is widened by using the chitosan immobilized enzyme. |
